Overview
Educational protocol for KPV research use, outlining study-based dosing for anti-inflammatory effects through once-daily subcutaneous injections.
What is KPV?
KPV (Lysine–Proline–Valine) is a tripeptide fragment derived from α-melanocyte-stimulating hormone (α-MSH). The peptide consists of the C-terminal three amino acids of this larger hormone, retaining potent anti-inflammatory properties without causing melanotropic side effects associated with full α-MSH.
Research demonstrates that KPV reduces pro-inflammatory cytokines in models of inflammatory bowel disease and systemic inflammation through inhibition of NF-κB signaling pathways.
How It Works
KPV operates by:
– Reducing pro-inflammatory cytokines (TNF-α, IL-6, IL-1β)
– Modulating immune cell activity
– Inhibiting nuclear factor kappa B (NF-κB) signaling
– Supporting inflammatory mediator regulation
– Providing systemic delivery through subcutaneous administration with rapid absorption
Reconstitution Instructions
Standard Approach (3 mL dilution):
1. Draw 3.0 mL bacteriostatic water with sterile syringe
2. Inject slowly down vial wall; avoid foaming
3. Gently swirl/roll until dissolved (do not shake)
4. Label and refrigerate at 2–8 °C, protected from light
Resulting Concentration: ~3.33 mg/mL
Measurement Conversion: At this concentration, 1 unit (U-100 insulin syringe) = 0.01 mL ≈ 33.33 mcg
Dosage Protocol
Gradual Titration Schedule (Recommended)
| Week | Daily Dose | Units (U-100 Syringe) | Volume (mL) |
|---|---|---|---|
| 1 | 200 mcg | 6 units | 0.06 mL |
| 2 | 300 mcg | 9 units | 0.09 mL |
| 3 | 400 mcg | 12 units | 0.12 mL |
| 4–8 | 500 mcg | 15 units | 0.15 mL |
Frequency: Once daily via subcutaneous injection
Cycle Duration: 8–12 weeks; optional extension to 16 weeks under monitoring
Administration: Inject at consistent time daily; rotate injection sites systematically
Storage Requirements
Lyophilized (Powder):
– Store at −20 °C (−4 °F) or below
– Maintain dry, dark conditions
– Protect from moisture and light
Reconstituted (Solution):
– Refrigerate at 2–8 °C (35.6–46.4 °F)
– Use within approximately 30 days
– Allow vials to reach room temperature before opening to minimize condensation
– Do not refreeze; avoid freeze-thaw cycles
Potential Benefits
Research observations include:
– “Anti-inflammatory activity: Reduces pro-inflammatory cytokines and modulates immune responses in models of inflammatory bowel disease and systemic inflammation”
– Oral and subcutaneous efficacy across multiple administration routes
– Preclinical data suggest support for tissue repair and wound healing processes
– Lack of melanotropic effects (no skin pigmentation changes)
Side Effects & Tolerability
Generally Well Tolerated:
– Occasional mild injection-site reactions (redness, slight swelling)
– Systemic side effects rarely reported in research protocols
– Local reactions can be managed with cool compress application
– Persistent reactions warrant protocol review
Prevention Strategies:
– Rotate injection sites at least 1–2 inches apart
– Inject slowly; wait seconds before withdrawing needle
– Use new sterile syringes for each administration
– Document daily dose, injection site, and observations
Supplies Needed (8-16 Week Protocol)
KPV Vials (10 mg each):
– 8 weeks: ~3 vials
– 12 weeks: ~4 vials
– 16 weeks: ~6 vials
Insulin Syringes (U-100):
– 8 weeks: 56 syringes
– 12 weeks: 84 syringes
– 16 weeks: 112 syringes
Bacteriostatic Water (10 mL bottles):
– 8 weeks: 1 bottle
– 12 weeks: 2 bottles
– 16 weeks: 2 bottles
Alcohol Swabs:
– 8 weeks: 2 x 100-count boxes
– 12 weeks: 2 x 100-count boxes
– 16 weeks: 3 x 100-count boxes
Injection Technique
1. Clean vial stopper and injection site with alcohol swabs; allow 10–15 seconds to dry
2. Pinch a 1–2 inch skinfold
3. Insert needle at 45–90° angle into subcutaneous tissue
4. Do not aspirate for subcutaneous injections
5. Inject slowly and steadily over 3–5 seconds
6. Withdraw smoothly; apply gentle pressure with clean swab (do not rub)
7. Rotate sites systematically using patterns (abdomen, thighs, upper arms)
8. Keep sites at least 1–2 inches apart
Preferred Injection Sites: Abdomen (2+ inches from navel), anterior/lateral thigh, outer upper arm
Lifestyle Optimization
Complementary strategies include:
– Anti-inflammatory diet emphasizing whole foods, omega-3s, polyphenols
– Minimize processed foods and refined sugars
– Stress management through meditation, yoga
– 7–9 hours of quality sleep nightly
– Regular moderate exercise (avoid overtraining)
– Gut health support through probiotics and adequate fiber
